Elimination of Mycoplasma Contamination in Cell Cultures

چکیده
These antibiotic treatments had a high efficiency of permanent cure: MRA 64%, ciprofloxacin 77%, BM-Cyclin 84%. Resistance to mycoplasma eradication was observed in some cell cultures: BM-Cyclin 5%, ciprofloxacin 14%, MRA 22%. Nearly all resistant contaminants that could be identified belonged to the species M. arginini and M. orale. Detrimental effects of the antibiotics were seen in form of culture death caused by cytotoxicity (in 9-13% of the cultures). Alterations of the cellular phenotypic features or selective clonal outgrowth might represent further untoward side-effects of exposure to these antibiotics. Overall, antibiotic decontamination of mycoplasmas is an efficient, inexpensive, reliable, and simple method: 269/366 (73%) chronically and heavily contaminated cultures were cured, while 97/366 (27%) cultures could not be cleansed and were either lost or remained infected. It is concluded that eukaryotic cell cultures containing mycoplasmas are amenable to antibiotic treatment and that a cure rate of about three-quarters is a reasonable expectation.

درمان آلودگی مایکوپلاسمایی در کشت سلول با سیپرو فلوکساسین و انروفلوکساسین: گزارش کوتاه
Background: Mycoplasma contamination in cell cultures is considered as a major economic, research and production problem. In this study, mycoplasma-infected Vero cell lines were treated by various dilutions of ciprofloxacin and enrofloxacin in a timely manner. Removal of mycoplasma contamination from infected cell cultures was evaluated and demonstrated by polymerase chain reaction (PCR) method...
متن کاملIsolation and detection of Mycoplasma pneumoniae from cell culture by culture and PCR.
Background: Mycoplasma pneumoniae is one of the major species of mycoplasmas which could contaminate cell cultures. Identification of M. pneuminae is significant because Mycoplasmas could contaminate and have unsuitable effect on the cell cultures. Rapid detection of these contaminations is so important and it could be significant role in preventing and controlling of contamination in cell cul...
متن کاملMycoplasma detection and elimination are necessary for the application of stem cell from human dental apical papilla to tissue engineering and regenerative medicine
BACKGROUND Recently, postnatal stem cells from dental papilla with neural crest origin have been considered as one of potent stem cell sources in regenerative medicine regarding their multi-differentiation capacity and relatively easy access. However, almost human oral tissues have been reported to be infected by mycoplasma which gives rise to oral cavity in teeth, and mycoplasma contamination ...
متن کاملThe prevalence of Mycoplasma hyorhinis contamination in tissues samples from cancer patients: A Brief Report
Infectious agents cause 15-20% of cancers worldwide. The infectious agents and contamination may be caused by a local chronic and advanced local inflammatory response, or by tumorization. Mycoplasma contamination can interfere with biological agents and cause DNA damage which affects gene expression, and disrupts the control of cell cycle inspection and apoptotic responses. Mycoplasmas are wide...
متن کاملThe Effects of Mycoplasma Contamination upon the Ability to Form Bioengineered 3D Kidney Cysts
Mycoplasma contamination of cell cultures is a pervasive, often undiagnosed and ignored problem in many laboratories that can result in reduced cell proliferation and changes in gene expression. Unless contamination is specifically suspected, it is often undetected in two dimensional (2D) cultures and the resulting effects of mycoplasma contamination are rarely appreciated and can lead to incor...
متن کاملElimination of Mycoplasma Contamination from Infected Human Hepatocyte C3A Cells by Intraperitoneal Injection in BALB/c Mice
Background/Aims: The use of antibiotics to eliminate Mycoplasma contamination has some serious limitations. Mycoplasma contamination can be eliminated by intraperitoneal injection of BALB/c mice with contaminated cells combined with screening monoclonal cells. However, in vivo passage in mice after injection with contaminated cells requires a long duration (20-54 days). Furthermore, it is impor...
